Not in the United States, but there are other professional leagues. Oh Sadaharu hit 868 home runs.

nsingman pointed out Sadaharu Oh. Josh Gibson (http://en.wikipedia.org/wiki/Josh_Gibson), who is estimated to have hit close to 800 in his Negro League career.
